Description

Quiescin sulfhydryl oxidase (QSOX) family, localized to the Golgi apparatus, catalyzes the insertion of disulfide bonds into a wide range of proteins by oxidizing sulfhydryl groups with the reduction of oxygen to hydrogen peroxide. QSOX family comprises of 2 proteins: QSOX1 and QSOX2. QSOX1 plays a role in protein folding, production of extracellular matrix, redox regulation, and protection from apoptosis, angiogenesis, and cell differentiation. QSOX1 forms a dimer upon cleavage of the C-terminal domain. It has two isoforms; isoform 1 (the full length of QSOX1) being expressed on Golgi apparatus membrane and isoform 2 (the splicing form) is secreted. QSOX1 activity is required for incorporation of laminin into the extracellular matrix (ECM) synthesized by fibroblasts, and ECM produced without QSOX1 is defective in supporting cell-matrix adhesion. This enzyme is efficiently secreted from mammalian cells despite the presence of a transmembrane domain (1).

QSOX1 is highly expressed in tumor cell lines but is not expressed in normal cells. In fibroblasts, it exhibits tumor-suppressing capabilities involved in growth regulation. The kinetics of QSOX1 expression portrays a more protective effect of QSOX1 rather than an involvement of this protein in apoptosis. Human breast cancer MCF-7 cell lines overexpressing the guinea pig QSOX1 protein appeared less sensitive to cell death than the MCF-7 control cells due to preservation of the mitochondrial polarization generally lost after an oxidative stress. QSOX1 is a potential new prognostic marker used in the staging of breast tumors and the stratification of breast cancer patients (2).The gene for QSOX1 is present on chromosome 1q24.

QSOX1- selective antibodies were generated against a peptide taken from the N-terminal region of the human protein.
